Semax raises a handful of sensible questions. This page answers them in order, starting with the fundamentals and moving to applications.
Reviewed 2025-09-06. Anything still debated is marked as such rather than presented as settled.
Development took place during the 1980s at institutes of the Russian Academy of Sciences, where peptide fragments of ACTH were screened for cognitive and neuroprotective effects. The compound received regulatory approval in Russia as a nasal preparation, marketed for conditions such as ischemic stroke, transient ischemic attacks, and optic nerve disorders. Registration in other countries has not followed. Most published clinical reports originate from a small number of Russian research groups, and independent replication outside that setting remains limited. The regulatory status therefore differs sharply between Russia and the rest of the world.
Laboratory work points toward modulation of neurotrophic signaling, particularly expression of brain-derived neurotrophic factor and nerve growth factor in hippocampal tissue. Studies also describe effects on monoamine turnover, inflammatory mediators, and oxidative markers. These observations come mainly from animal models and cultured cells, so the causal chain in humans is not firmly established. Whether the reported molecular changes translate into measurable clinical benefit is an open question. Reviews generally present the mechanism as plausible rather than demonstrated.

Reported pharmacological work centers on neurotrophic signaling, including changes in BDNF and NGF expression in hippocampal tissue in animal models. Human data come largely from studies conducted in Russia, and how well those results generalize to other populations remains an open question. Regulatory status differs sharply by jurisdiction: Semax is a registered prescription medicine in Russia, while it holds no approved marketing status in the United States or the European Union. Outside such jurisdictions it is generally handled as a research chemical, which affects both documentation and quality expectations.
Semax is a synthetic heptapeptide whose sequence is Met-Glu-His-Phe-Pro-Gly-Pro. It was developed as a fragment analog of adrenocorticotropic hormone, modeled specifically on the ACTH(4-10) region. The first four residues reproduce that fragment, while a Pro-Gly-Pro tripeptide is appended at the C-terminus. Work on the compound originated in Russia, where it entered clinical use as an intranasal preparation. Its sequence places it among short regulatory peptides studied for effects on the central nervous system rather than on the adrenal axis.
The C-terminal Pro-Gly-Pro extension is not incidental. Proline-rich tails are known to resist several common peptidases, and the published literature attributes the longer half-life of Semax, relative to unmodified ACTH fragments, to this feature. The modification also removes the melanocyte-stimulating and corticosteroidogenic activity that characterizes longer ACTH-derived sequences. Because the molecule is small and hydrophilic, it is typically formulated as an aqueous solution for intranasal or parenteral delivery. Acetylation or amidation at the termini appears in closely related research peptides and shifts the mass by a fixed increment.
== Legacy == Duttaroy's research focuses on how the placenta transports maternal plasma DHA and ARA to the fetus. The fetal brain development in utero is critically dependent on the maternal supply of these fatty acids in the utero. His studies have been documented through several articles. Besides, his articles have drawn many citations (Google Scholar PubMed). His other research area is cardioprotective factors in fruits and vegetables. He has discovered the anti-platelet factors from tomatoes. This discovery made by him is now widely known as Fruitflow. In 2009, Fruitflow® became the first product in Europe to obtain an approved, proprietary health claim under Article 13(5) of the European Health Claims Regulation 1924/2006 on nutrition and health claims made on foods. The EU Commission authorized the health claim "water-soluble tomato concentrate (WSTC) I and II help maintain normal platelet aggregation, which contributes to healthy blood flow." Fruitflow® is now widely available in different countries worldwide.
A U.S. company named VAZTEC has created a compliant rotary valve sealing system that solves the friction and sealing problems of previous designs. They have built ten prototype engines from a 5.3L V-8 to a handheld 28cc four stroke. VAZTEC has also built a successful Diesel engine that contained the high compression pressures of a compression ignition engine (20:1 compression ratio, 100 bar combustion pressure). They are working with various OE manufacturers to commercialize their design. Various patents cover the compliant seal, including U.S. Patent 9,903,239. A Computational Fluid Dynamics (CFD) model of the VAZTEC rotary valve can be viewed at this link.

Plants are a rich source of ribosomally-synthesised and post-translationally modified cyclic peptides. Among these, the cyclotides are gene-coded products generated via processing of a larger precursor protein. The gene for the first such precursor is Oak1 (Oldenlandia affinis kalata clone number 1), which was shown to be responsible for the synthesis of kalata B1. The generic configuration of the precursor protein consists of an endoplasmic reticulum signal sequence, a non-conserved pro-region, a highly conserved region known as the N-terminal repeat (NTR), the mature cyclotide domain and finally a short hydrophobic C-terminal tail. The cyclotide domain may contain either one cyclotide sequence, as in the case of Oak1, or multiple copies separated by additional NTR sequences as seen for Oak2 and Oak4. In precursor proteins containing multiple cyclotide domains these can either be all identical sequences, as is the case for Oak4, or they can be different cyclotides as in Oak2 which contains sequences corresponding to kalata B3 and B6. Recently, the enzyme responsible for the backbone cyclization of cyclotides has been isolated from the medicinal plant Clitoria ternatea. This enzyme was named butelase 1 in accordance to the local name of the plant (Bunga Telang Ligase). Butelase 1 has been shown to cyclize the linear precursor of kalata B1 with >95% yield at a remarkable rate of 5.42×105 M−1 s−1.

=== Derivatives === A number of derivatives of 18-MC have been developed, with several of them being superior to 18-MC itself, the methoxyethyl congener ME-18-MC being more potent than 18-MC with similar efficacy, and the methylamino analogue 18-MAC being more effective than 18-MC with around the same potency. These compounds were also found to act as selective α3β4 nicotinic acetylcholine antagonists, with little or no effect on NMDA receptors.
An anorectic is a drug that reduces appetite, resulting in lower food consumption, leading to weight loss. These substances work by affecting the central nervous system or certain neurotransmitters to create a feeling of fullness or reduce the desire to eat. The understanding of anorexiant effects is crucial in the development of interventions for weight management, eating disorders, and related health concerns. The anorexiant effect can be induced through diverse mechanisms, ranging from hormonal regulation to neural signaling. Ghrelin, leptin, and peptide YY are among the hormones involved in appetite control. Additionally, neurotransmitters such as serotonin and dopamine in the central nervous system contribute significantly to the regulation of food intake. By contrast, an appetite stimulant is referred to as orexigenic. The term is (from the Greek ἀν- an- 'without' and ὄρεξις órexis 'appetite'), and such drugs are also known as anorexigenic, anorexiant, or appetite suppressant.
In 1960, the Grace Line, inspired by the pioneering efforts of Sea-Land Service, Matson Navigation, and Seatrain Lines, sought to begin containerizing its South American cargo operations by converting the conventional freighters Santa Eliana and Santa Leonor into fully cellular container ships. The effort was stymied by the opposition of longshoremen in New York and Venezuela, and the ships were repeatedly laid up idle and were ultimately sold to the domestic container line Sea-Land Service in 1964. In 1963 Grace made a second attempt to containerize its South American trade when it ordered the four M-class combination passenger-cargo ships Santa Magdalaena, Santa Maria, Santa Mariana, and Santa Mercedes with partial cellular holds, but they were no more successful as mixing conventional break-bulk cargo and containers in the same ship negated the operating economies that full containerization promised. In 1970, Grace Line was sold to Prudential Lines for $44.5 million, with the merged company renamed Prudential Grace Line. It was taken over by Delta Steamship Lines in 1978, thereby extinguishing the name Grace in ocean shipping. Subsequently, Delta Steamship Lines was acquired and consolidated by Crowley Maritime in 1982.
However, because of how quickly their revolts would further radicalize the lower classes, the Creoles quickly stopped supporting general violent insurrection because they benefitted from social change that occurred through the systems of the Spanish crown. Institutional change ensured stability by supporting the political institutions that allowed for the creation of a wealthy Creole class and further adapting those institutions to meet demands, rather than propose a radical shift in the complete make-up of socioeconomic life and traditions. However, institutional change did not come as anticipated and further spurred on the radicalization of Spanish-American social classes towards independence.
The western regions of Pakistan became a part of the Achaemenid Empire around 517 BCE. In 326 BCE, Alexander the Great conquered the region by defeating various local rulers, most notably, the King Porus, at Jhelum. Among the major powers that ruled the region were the Mauryas (322–185 BCE), during which Ashoka the Great extended the empire. The Indo-Greek Kingdom founded by Demetrius of Bactria (180–165 BCE) included Gandhara and Punjab and reached its greatest extent under Menander (165–150 BCE), allowing the Greco-Buddhist culture in the region to prosper. Taxila had one of the earliest universities and centres of higher education in the world, which was established during the late Vedic period in the 6th century BCE. The ancient university was documented by the invading forces of Alexander the Great and recorded by Chinese pilgrims in the 4th or 5th century CE. At its zenith, the Rai dynasty (489–632 CE) ruled Sindh and the surrounding territories.

The Royal Air Force (RAF) provided surveillance and reconnaissance support, with an RAF surveillance plane tracked over the area where a Russian-linked tanker, Bella 1, was seized by US forces in January 2026. The UK provided one of its largest naval supply ships, the RFA Tideforce to support the US seizure of Bella 1 (renamed Marinera) in the North Atlantic, and UK military bases in the Atlantic were used by US planes and military assets involved in the operation.
==== Coupling with separation techniques ==== DART can be combined with many separation techniques. Thin-layer chromatography (TLC) plates have been analyzed by positioning them directly in the DART gas stream. Gas chromatography has been carried out by coupling gas chromatography columns directly into the DART gas stream through a heated interface. Eluate from a high-pressure liquid chromatograph (HPLC) can be also introduced to the reaction zone of the DART source and analyze. DART can be coupled with capillary electrophoresis (CE) and the eluate of CE is guided to the mass spectrometer through the DART ion source.
== History == Dimethocaine was originally synthesized by the Hoffmann-La Roche company in 1930. It was sold under the market name larocaine. During the 1930s dimethocaine gained popularity in the US as a local anesthetic. Just like cocaine and procaine, it was used during surgery, primarily in dentistry, ophthalmology and otolaryngology. However, in the 1940s, it was removed from the market because of its psychoactive effects and risk of addiction. Nowadays dimethocaine is abused for these psychoactive effects. It is sold as a cocaine surrogate to circumvent legislation issues.
=== Neuroprotective activity === In vitro studies have demonstrated that didymin can rescue neuronal cells from oxidative damage. In a membrane-based neuronal cell model, it inhibited hydrogen peroxide-induced mitochondrial dysfunction, reduced caspase-3 activation, and suppressed JNK phosphorylation, suggesting potential relevance for neurodegenerative diseases associated with oxidative stress.

Semax is a synthetic seven-amino-acid peptide derived from a fragment of corticotropin. It is used in Russia as a nasal preparation, while elsewhere it is studied as a research compound. It is not approved as a medicine in most countries.
Its sequence corresponds to the 4-10 region of corticotropin extended by three residues. This fragment lacks the region responsible for adrenal steroid release, so it is explored for neural rather than hormonal effects. The relationship explains the compound's classification among ACTH-derived peptides.
It holds approval in Russia for several neurological indications as a nasal spray or drop formulation. Regulatory agencies in the United States and the European Union have not approved it. Products sold elsewhere are generally labeled for research use only.
