=== Forcing association of protein complexes === As an example, emicizumab (formerly RG6013) is an IgG derivative containing H-chain heterodimerization motifs, which was combined with the common light chain approach to prevent L-chain mispairing issues. With a bivalent composition, emicizumab brings two protein antigens together into one complex. Factor IXa and Factor X in the coagulation cascade are the cognate antigens which are bound by RG6013. These two factors are brought together by coagulation factor VIIIa in a healthy individual, while patients with bleeding disorder hemophilia A do not have VIIIa. Current treatment of this disorder is to supplement the patients with FVIII to reduce bleeding complications. But FVIII can be recognized as a foreign protein in these patients due to the absence of this protein and thus an immune response will be generated against this protein. Besides, FVIII has a short half-life (less than 15 hours) and thus is cleared rapidly. === Synthetic === The availability of synthetic toxin has allowed scientists to further explore the biological activity of the toxin, resulting in the observation that d-ACTX-Ar1a causes repetitive firing and prolongation of the action potential. These actions underlie the clinical symptoms seen following envenomation and further contribute to the understanding of the molecular basis for activity of this potent neurotoxin on voltage-gated sodium channels. Under voltage-clamp conditions in dorsal root ganglion (DRG) neurons it was found that the effects of the synthetic toxin on sodium currents were not significantly different from those previously reported for the native toxin. Neither native nor synthetic d-ACTX-Ar1a had any effect on TTX-resistant sodium currents, but both exerted a potent selective modulation of TTX-sensitive sodium currents consistent with actions on neurotoxin receptor site-3. This includes a slowing of the sodium-channel inactivation, a hyperpolarizing shift in the voltage-dependence of activation and a hyperpolarizing shift in the steady-state sodium-channel inactivation. d-ACTX-Ar1a causes a prolongation of action potential duration, accompanied by spontaneous repetitive firing, but does not depolarize the resting membrane potential. Effects on the autonomic nervous system, including vomiting, profuse sweating, salivation, lachrymation, marked hypertension followed by hypotension, together with effect on the somatic nervous system to cause muscle fasciculation and dyspnea (shortness of breath) are presumably due to excessive transmitter release.

Each freeze-thaw cycle can degrade a fraction of the peptide and promote aggregation. Dividing a stock solution into single-use volumes removes repeated cycling as a variable. It also limits the time a thawed solution spends at room temperature.
A high main-peak percentage says little about identity, counter-ion content, or residual solvents. It also does not distinguish an isomer or a closely eluting analog. Mass confirmation and the accompanying documentation cover those gaps.
Salt form changes mass, solubility, and hygroscopicity, and acetate and trifluoroacetate salts behave differently in solution. Storage guidance still centers on -20 °C for powder and lower temperatures for solutions. The certificate should state which salt is present.
