===== Jovita Rivera ===== Jovita Rivera was one of the ten plaintiffs in the federal class action suit of Madrigal v. Quilligan. On October 12, 1973, Rivera went to the USC-LA hospital to give birth to her second child. She was under medication and in labor pains when medical staff misinformed her about the risk and chances of getting pregnant right after birth. She consented and a tubal ligation was done. Rivera, 27 at the time, states that during her stay at the hospital, while in advanced labor and under pain medication due to complications, her doctor told her she would be a burden to the government. Women like Rivera were offered the choice of sterilization under poor circumstances, under medication, and with no language assistance for translation. Some of the other plaintiffs for this case faced hostility from staff when told they could receive more pain medication if they signed papers consenting to sterilization. Rivera stated: "... the doctor told me that I had too many children, that I was poor, and a burden to the government and I should sign a paper not to have more children [...] The doctors told me that my tubes could be untied at a later time and I could still have children." While Rivera was under distress, she believed the process was reversible and consented. When Rivera and the other plaintiffs testified in court to prove that they had been coerced into getting a procedure, the judge did not rule in their favor.
==== Preclearing ==== Lysates are complex mixtures of proteins, lipids, carbohydrates and nucleic acids, and one must assume that some amount of non-specific binding to the IP antibody, Protein A/G or the beaded support will occur and negatively affect the detection of the immunoprecipitated target(s). In most cases, preclearing the lysate at the start of each immunoprecipitation experiment (see step 2 in the "protocol" section below) is a way to remove potentially reactive components from the cell lysate prior to the immunoprecipitation to prevent the non-specific binding of these components to the IP beads or antibody. The basic preclearing procedure is described below, wherein the lysate is incubated with beads alone, which are then removed and discarded prior to the immunoprecipitation. This approach, though, does not account for non-specific binding to the IP antibody, which can be considerable. Therefore, an alternative method of preclearing is to incubate the protein mixture with exactly the same components that will be used in the immunoprecipitation, except that a non-target, irrelevant antibody of the same antibody subclass as the IP antibody is used instead of the IP antibody itself.

==== 2012 Jalisco massacres ==== The dismembered remains of 18 bodies were found inside a Toyota Sienna and Ford EcoSport near the U.S. retiree communities in Chapala, Jalisco, just south of the city of Guadalajara. Eighteen heads were found along the dismembered bodies; some had been frozen, others were covered in lime, and the rest were found in an advanced state of decomposition. An anonymous call alerted the police to the abandoned vehicles, which were found by the side of a highway early in the morning on 9 May 2012. They were consequently towed to government offices to unload the bodies. The authorities confirmed that a message was left behind by the killers, presumably from Los Zetas and the Milenio Cartel. The attorney general of the state of Jalisco, Tomás Coronado Olmos, stated that this massacre was a revenge attack for the 23 killed in the 2012 Nuevo Laredo massacres. In addition, 25 people were rescued after being kidnapped in Tala, Jalisco on 8 May 2012; the killers had plans to kill and "throw" them for public display. Another 10 people managed to escape their capture by members of Los Zetas that same day, and alerted the local media of the situation. Upon the arrest of the four alleged killers, one of the cartel members confessed that they had plans to "repeat" what had happened in the 2011 Guadalajara massacre, where 26 bodies were dumped in a major avenue for public display. According to Proceso magazine, Los Zetas were planning to kill 50 people on 9 May 2012, a day before Mother's Day.

=== Phase transitions === The melting point of crystallized honey is between 40 and 50 °C (104 and 122 °F), depending on its composition. Below this temperature, honey can be either in a metastable state, meaning that it will not crystallize until a seed crystal is added, or, more often, it is in a "labile" state, being saturated with enough sugars to crystallize spontaneously. The rate of crystallization is affected by many factors, but the primary factor is the ratio of the main sugars: fructose to glucose. The next day, the restaurant chain reopened 50 more restaurants in Moscow and the wider Moscow region. McDonald's has a 15-year option to buy its former restaurants back from Vkusno i tochka. === Murine === Initial therapeutic antibodies were murine analogues (suffix -omab). These antibodies have: a short half-life in vivo (due to immune complex formation), limited penetration into tumour sites and inadequately recruit host effector functions. ==== Base-specific cleavage/MALDI-TOF ==== A recently described method by Ehrich et al. further takes advantage of bisulfite-conversions by adding a base-specific cleavage step to enhance the information gained from the nucleotide changes. By first using in vitro transcription of the region of interest into RNA (by adding an RNA polymerase promoter site to the PCR primer in the initial amplification), RNase A can be used to cleave the RNA transcript at base-specific sites. As RNase A cleaves RNA specifically at cytosine and uracil ribonucleotides, base-specificity is achieved by adding incorporating cleavage-resistant dTTP when cytosine-specific (C-specific) cleavage is desired, and incorporating dCTP when uracil-specific (U-specific) cleavage is desired. The cleaved fragments can then be analyzed by MALDI-TOF. Bisulfite treatment results in either introduction/removal of cleavage sites by C-to-U conversions or shift in fragment mass by G-to-A conversions in the amplified reverse strand. C-specific cleavage will cut specifically at all methylated CpG sites. By analyzing the sizes of the resulting fragments, it is possible to determine the specific pattern of DNA methylation of CpG sites within the region, rather than determining the extent of methylation of the region as a whole. This method demonstrated efficacy for high-throughput screening, allowing for interrogation of numerous CpG sites in multiple tissues in a cost-efficient manner.
== Diagnosis == Substance use disorders can be confused with other psychiatric disorders.
